Obesity is a disease affected by many factors including exercise. Adipose tissue is exposed to various types of mechanical forces such as compression, tension and shear stress. Moreover, exercise leads to increased levels of mechanical stimulation on adipose tissue and cell. Therefore, understanding the adipocyte response to mechanical stimulation is important in studies of obesity. In this study, we investigated the effects of fluid shear stress on adipogenic differentiation. Preadipocytes were induced to differentiate with induction medium containing dexamethasone, 3-isobutyl-1-methylxanthine and insulin on day 0. Dynamic fluid shear stress of maximum 1 Pa at 1 Hz was applied for 1 hour at either pre-differentiation (day 0) or post-differentiation (day 5). Fluid shear stress was not applied in control group. Adipocyte differentiation-related proteins were assessed by Western blot analysis. Glycerol-3-phosphate dehydrogenase (GPDH) activity, a marker of adipocyte differentiation, was determined by GPDH assay and formation of lipid droplets were stained with Oil Red O. CCAAT/enhancer-binding protein (C/EBP) beta, C/EBP alpha, peroxisome proliferator-activated receptor (PPAR) gamma which induce adipogenic differentiation were reduced by fluid shear stress. GPDH activity also decreased in mechanical loading groups. The decrease in the number of lipid droplets was enhanced in post-differentiation loading group. Results from this study suggest that fluid shear stress inhibits adipocyte differentiation especially when fluid shear stress is applied at post-differentiation.
