To develop a useful diagnostic tools for detecting the cattle with potential for developing leucosis from the BLV antibody positive cattle, monoclonal antibodies(Mabs) reacted with tumor-associated antigens(TAA) of bovine leucosis were prepared, and efficacy of detecting TAA in the lymphocytes from leukotic cattle was evaluated by using the Mabs. One hundred and thirty one cattle were examined by hematological test, agar gel immunodiffusion(AGID) test and indirect immunofluorescent(IFA) test for enzootic bovine leucosis. The positive rates were 9.9% by hematology and 21.4% by AGID. Among the positive, four cattle showing BLV-positive, persistent lymphocytosis and TAA positive could be selected. These cattle showed notably increased number of surface immunoglobulin-bearing cells compared with the number of rosette forming cells in peripheral blood. Six Mabs specifically reacted with TAA in the lymphocyte from the known TAA-positive cattle were obtained. Antibody titers of the Mabs in the ascitic fluids of mice and cell culture fluids were 32 to 256 and 2 to 8 by IFA and 512 to 2048 and 32 to 128 in complement dependent cytotoxicity (CDAC) test, respectively. Immunoglobulin isotypes of the Mabs were classified as IgG1, IgG2a, IgG2b and IgM. Molecular weights of TAA for the Mabs measured by Western blots were 65K, 74K, 95K, 136K and 174K. By using the Mabs, TAA positive rates of the field samples from healthy BLV-positive cattle were found to be 87.0% by IFA and 82.6% by CDAC. Korean black goats at the 24th weeks post inoculation of BLV showed 85.7 % of BLV antibody-positive and 14.3 /o and 42.9% of TAA positive by IFA and CDAC respectively.
