The present study was carried out to investigate the potential acute toxicity of new enzyme-treated royal jelly (RJ) by a single oral dose in Sprague Dawley (SD) rats. The sample was prepared by treating the RJ with four enzymes approved as food additives by the KFDA for improving digestion and absorption of nutrients. Male and female rats were randomly assigned to four groups of 10 rats each. The test articles were administered once by the gavage to rats at dose levels of 0 (placebo control), 1,250, 2,500 and 5,000mg/kg body weight. Mortalities, clinical findings, and body weight changes were monitored for 14 days after dosing. At the end of 14 day observation period, all animals were sacrificed, and the completed gross postmortem examinations were performed. There were no dead animals and specific adverse effects on clinical signs, the body weight, and the gross finding on males and females of the all groups, except for the transient clinical signs such as polyuria and diarrhea. Consequently, no toxic effect was observed at the dose levels of 1,250, 2,500, or 5,000mg/kg body weight in rats. The approximate lethal dose (ALD) of new enzyme-treated freeze dried (ETFD) RJ may be more than 5,000mg/kg body weight. Therefore, oral administration of ETFD RJ did not show any toxic effects to all rats upto 5,000mg, the highest dosage tested in this study and is likely to be safe in humans.
