The present study was performed to investigate the effect of an intensive endurance exercise on muscle protein synthesis in aged skeletal muscle. Young (12 wk) and aged (l00 wk) male Sprague-Dawley rats (N=56) were randomly assigned to the time course of the treadmill running (75-80% VO₂ max, 1hr). Muscle samples were obtained before and during the recovery period of the running (immediately after, 1hr, and 3hr after the running) from EDL muscle. PKB/Akt, mTOR, p70S6k, 4E-BP1 and AMPK phosphorylation were determined by SDS-PAGE and western blotting. From the results of this study, we confirmed Akt(298%, p=.001), mTOR(389%, p=.001) and p70S6K(397%, p=.009) in young group, as well as Akt(l73%, p=.001), mTOR(193%, p=.001) and p70S6K(335%, p=.001) in old group were significantly increased after the intensive endurance running (p<.05). These increased responses were maintained up to 3 hours of the recovery in both groups. The level of p70S6K phosphorylation was higher in young than old group (35%, p=.002), on the contrary to the level of AMPK phosphorylation was higher in old than young group (25%, p=.001). These results suggest that intensive endurance exercise positively effect on muscle protein synthesis not only in young but also in aged skeletal muscle.
