A novel loop-mediated isothermal amplification (LAMP)-based detection method was established for the rapid detection of Paenibacillus larvae (P. larvae), which is the causative agent of American foulbrood (AFB). This method enabled amplification of a part of the metalloproteinase (Mtpr) gene at a temperature of 63℃ within 45min, with no need for thermal cycling. The specificity of the method was demonstrated by successful amplification of the bee genome infected with P. larvae and by HindⅢ restriction enzyme analysis applied to the inner primers. The present method detected as low as 2 fg of DNA plasmid. Visual inspection of LAMP products using Gene finder demonstrated that the positive and negative reactions exhibited distinct and different colors in daylight to the naked eye without a UV illuminator or other supporting tools, which means that gel electrophoresis was not necessary to appreciate the presence of P. larvae. Therefore, this detection method is rapid, sensitive, specific, and low in cost and it may be used to facilitate development of apiaries.
