To establish the efficient in vitro propagation of Sedum sarmentosum, the capacities of callus induction and plant regeneration were compared in different basal medium (B5 and Murashige and Skoog, MS), explant source (leaf and stem) and collection (28 locals). Plant regeneration was more intensive in MS medium as compared to the B5, and plantlets were produced through shoot organogenesis. Leaf explant was more responsive in callus induction and plant regeneration compared to the stem. Callus induction from leaf explants showed the high frequency range (84-100%) in all 28 collections, and successful plant regeneration occurred in only 10 collections. Keumsan collection showed the highest plant regeneration ability (100%, 16.3 plantlets/explant) followed by Bucheon (96.7%, 11.8 plantlets/explant) and Pocheon (90.0%, 4.2 plantlets/explant) at 50 days after transferring calluses. The callus induction and plant regeneration of S. sarmentosum were largely affected by genotype, explant source and basal medium, and the plant regeneration efficiency appeared to be the result of an interaction among these factors.
