Repeated dose toxicity of sodium chlorate was evaluated according to the Toxicity Test Guidelines of the Korea Food and Drug Administration using ICR mice. Groups of 10 male and 10 female ICR mice were exposed to drinking water containing 0, 37.5, 75, or 150 mM sodium chlorate for 4 weeks. All mice survived to the end of the study. Mean body weights of the exposed male and female mice were similar to those of controls. Feed and water consumption of the exposed and control groups was similar. No gross lesions related to sodium chlorate exposure were observed. In the hematological analysis of the male group treated with 150 mM sodium chlorate, white blood cells, neutrophils, and eosinophils were significantly decreased, and mean corpuscular hemoglobin concentration and platelets were significantly increased. In the treated female group, the concentration of hemoglobin was significantly increased and that of platelets was significantly decreased. In blood biochemical analysis of mice treated with 150 mM sodium chlorate, concentration of AST, ALT, and creatinine in both the male and female groups were significantly different compared with the control group. In mice treated with 150 mM sodium chlorate, the histopathological kidney findings of the treated mice showed severe necrosis and vacuolation of cells in convoluted tubules, and the histopathological liver findings of the treated mice showed severe necrosis and depletion of nuclei. The significant differences in the hematological and blood biochemical analyses and abnormal histopathological findings in the mouse kidneys and livers may have been related to sodium chlorate exposure. From the results of this study, the no observed adverse effect level (NOAEL) of sodium chlorate was approximately 37.5 mM in both male and female mice.
