Exercise is a commonly used therapeutic procedure in the management for patient of Parkinson's disease; But, neuronal changes with exercise for patients with Parkinson's disease has not been well established, although some clinical and laboratory reports suggest that regular exercise may produce a neuroprotective effect and restore dopaminergic and motor functions. The purpose of this study was to determined the effects of treadmill exercise on the nigrostriatal dopaminergic neurodegeneration in MPTP induced mouse model of Parkinson Disease.
For this study, we used a chronic mouse model(male C57BL/6 mice) of Parkinsonism, which was induced by injecting with 10 doses(Every 12 hour) of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) 30 ㎎/㎏ and probenecid (20 ㎎/㎏) over 5 days. At the end of exercise training, we examined and compared their locomotor ability(behavior test), neuronal and neurochemical changes with group of control parkinsonian(No-exercise)mice.
The results of this study were summarized as follows:
1) Treadmill training decreased synphilin expression levels in substantia nigra and striatum of MPTP-induced parkinson's disease model.
2) In MPTP-induced parkinson model, treadmill exercise inhibits decrease of cell numbers in substantia nigra and striatum.
3) In MPTP-induced parkinson model with dopamine treatment, treadmill exercise increase p-ERK1/2 expression levels in substantia nigra.
4) The locomotor ability(behavior test) after 4 weeks of exercise in the chronic parkinsonian mice group was significantly higher rate than the pre-exercise, whereas the locomotor ability(behavior test) at the beginning and 4 weeks afterward in the control(No-exercise) parkinsonian mice was unchanged.
The results of this study suggest that treadmill exercise partially prevented dopaminergic neurodegeneration in the nigrostriatal(substantia nigra, striatum, dentate gyrus) region of the Parkinson's Disease model, and this effects were may offer a potential therapeutic adjunct to current Parkinson's Disease therapies in the future.
