This study was conducted to establish mating procedure, pregnancy diagnosis and Caesarean section in Cynomolgus monkeys. Six males, aged 6~7 years, weighing 4~7 ㎏, and 18 females showing regular menstrual cycle, aged 4.5~6 years, weighing 3~5.6 ㎏ were selected. Mating was conducted daily for three consecutive days among 11 to 15 days after the menstruation start. As to mating, we established
the natural mating method, which cohabited male and female monkeys in a cage and produced mated animals. As a result of three days’ natural mating, 15 out of 18 female animals (83%) succeeded in mating, and 8 out of the mated ones (53%) succeeded in pregnancy. Mating was confirmed through observing the mating behavior and checking sperm in vagina smear. Pregnancy diagnosis and observation of embryonic development were performed using ultrasonic diagnostic equipment. According to the results of the tests, the endometrial cavity was opened by around a third on Day 18 of pregnancy, and the egg yolk and heart rate of the endometrial cavity were observed from Day 25. The fetal form was observed to some degree on Day 35, and the completed form of fetus was observed on Day 45. Caesarean section was conducted on Day 50 of pregnancy. To anesthetize pregnant animals, we performed intramuscular injection of tranquilizer, which was 0.1 mg/kg of medetomidine. We also applied intramuscular injection of 10 mg/kg of ketamine as an anesthetic and maintained oxygen and 1-2% of isoflurane using a mask to maintain anesthesia during operation. The condition of the gravid uterus, the survival and the external examination of the fetus were observed. In conclusion, the natural mating method established in this study was found to be a useful tool for obtaining pregnant dams and for evaluation of reproductive toxicity and risk assessment of new drug candidates.
