Pulsed radiofrequency (PRF) stimulation into dorsal root ganglion (DRG) has been recently introduced as an alternative method of pain intervention in patients with neuropathic pain. Animal studies further reveal that PRF stimulation into DRG increases Fos expression (neuronal activity marker) in the spinal cord. Thus, it is assumed that PRF induced neuronal activation may be closely related with long-term analgesic effect in the neuropathic conditions. Presently, it is not clear PRF application into peripheral nerve fiber including sciatic nerve also produces analgesic effect in neuropathic pain condition. We firstly evaluated whether PRF application for two minutes into right sciatic nerve of rat in mid thigh increased spinal Fos expression. And then we further investigated whether this PRF application produced analgesic effect on the neuropathy using rat chronic constriction injury (CCI) model. PRF application did not significantly increase Fos expression in the spinal cord as compared with control animals. Furthermore, PRF application failed to reduce the mechanical allodynia and non-noxious stimulation induced spinal Fos expression in CCI model. Therefore, this data suggest that PRF treatment into sciatic nerve do not produce the spinal neuronal activity as well as pain relief in neuropathic animal model.
