Cultivated Daphne odora plants showing chlorosis and stunting were observed and their causal agent was identified and molecular detection method was developed. An isometric virus was isolated from infected leaf tissues of some collected Daphne plants and partial region of genome RNA2 covering the coat protein gene for the virus was cloned for sequence determination to identify the virus. The sequence analysis suggests that the virus was identified as an isolate of Cycas necrotic stunt virus (CNSV-Do1). Reverse transcription-polymerase chain reaction (RT-PCR) was developed to detect the virus. RT-PCR was performed to screen disease incidence of CNSV in Daphne plants, and 13 out of 17 D. odora plants (76.5%) were infected by CNSV. This study suggests that CNSVspecific primer set can specifically detect the virus and can be useful for indexing of the virus-free Daphne plants.
