本 硏究는 모자이크 病徵을 나타내는 쑥부쟁이(Aster yomena Makino) 잎에서 分離한 오이 모자이크 바이러스(cucumber mosaic virus, CMV-As)의 特性을 調査하기 위하여 實施되었다. CMV-As의 增殖寄主를 담배(Nicotiana tobacum cv. Xanthi-nc)를 使用하여 바이러스를 增殖시켰다. CMV-As의 物理的 性質을 동부에 接種試驗 및 寒天 젤 擴散法을 使用하여 調査한 結果, 耐熱性이 65℃, 耐稀釋性이 10³, 그리고 耐保存性이 3-4日이었다. 蟲媒傳染試驗 結果, 복숭아혹진딧물이 CMV-As系統의 媒介體이며 約 30%의 바이러스 傳染率을 보였다. 純化된 CMV-As系統의 紫外線 吸收樣相을 調査한 結果 260 ㎚에서 最大, 240 ㎚에서 最小致의 바이러스 樣相을 보였으며, 最大置와 最小置의 比 A260/A280 및 A280 A260의 比는 각각 1.27, 1.76, 그리고 0.57이었다. A260/A280 比를 土臺로 할때 CMV-As內 RNA 含量이 18%임을 推定할 수 있었다. 純化된 CMV-As를 電子顯微鏡으로 檢鏡한 結果, 直徑이 約 30㎚인 球形이며, 粒子 中心에 hollow center를 가진 전형적인 CMV 粒子를 觀察할 수 있었다. 電氣泳動(SDS-PAGE)으로 CMV-As 外皮蛋白質을 構成하는 小單位의 種類 및 分子量을 調査한 結果, 約 25,000 dalton의 單一 蛋白質밴드를 確認하였으며, 이는 CMV-As 外皮蛋白質을 構成하는 小單位의 種類가 1種으로 이뤄졌음을 意味한다. 포름아미드가 包含된 agarose젤 電氣泳動으로 檢定한 結果, 4種의 게놈 RNA와 1개의 衛星 RNA가 分離되었으며 이들의 分子量은 各各 1.29, 1.16, 0.74, 0.34 및 0.136×10?dalton으로 推定되었다.
This study was conducted to characterize an isolate of cucumber mosaic virus(CMV-As) from Aster yomena Makino showing mosaic symptoms on the leaves. The virus was multiplied in the leaves of Nicotiana tabacum cv. Xanthi-nc. Physical properties of CMV-As, determined by inoculations on Vigna sinensis and the Ouchterlony's agar gel double diffusion test, were as follows: thermal inactivation point was 65℃, dilution end point was 10?³, and longevity in vitro was 3-4 days. The green peach aphid(Myzus persicae was a vector of CMV-As and the percentage of transmissions was about 30%. Ultraviolet absorption pattern of purified CMV-As indicated a maximum at 260㎚, and a minimum at 240㎚. The ratios of Amax Amin, A260 A280, and A280 A260 were 1.27, 1.76, and 0.57, respectively. Based upon the ratio of A260 A280, RNA content of CMV-As was estimated to be 18%. Electron micrograph showed that the purified CMV-As was isometric particle about 30㎚ in diameter with hollow center. Sodium dodecyl sulfate(SDS)-polyacrylamide gel electrophoresis(PAGE) was adopted for determination of the molecular weight and the number of subunit of CMV-As coat protein ; molecular weight was about 25,000 daltons, and one major protein band was shown, which indicated that the virus had one type of subunit. Formamide-agarose gel electrophoresis showed that CMV-As consisted of four genomic RNAs(RNA 1, 2, 3, 4) and a satellite RNA. Estimated molecular weights of RNAs were 1.29, 1.16, 0.74, 0.34, and 0.136×10? daltons, respectively.
