This study was carried out to analysis the genetic characteristics and to find out breed specific genetic markers of Korean Native Chicken. For those purpose, 1) general performances in economic traits(body weight(BW270), first egg days(SM), egg weight at 270 days of age(EW270) and egg production(EP270)) were analysed ; 2) the eight biochemical polymorphic loci controlling blood proteins(Tf, Hb, Pol, Al) and enzymes(Cat, Am-1, Es-l, Akp) were analysed and 3) polymorphic DNA markers were screened in Korean Native Chicken, classified on the basis of the feather color and appearance (Korean Native Red Chicken(KNRC), Korean Native Brown Chicken(KNYC), Korean Native Black Chicken(KNBC)).
In growth rate, KNBC was higher than any other 2 strains(KNYC, KNBC). Difference of growth rate between female and male was observed after the 8 weeks of age and body weight at 22 weeks of age showed 1,979g. Overall means of economic traits(3 strains) were 147.3day for the days of first eggs, 49.5g for EW270, 62.8% for EP270, and 1,889g for BW240.
In analysis of genetic characteristics using the biochemical markers in Korean Native Chicken, the frequencies of B and C alleles for Tf locus were 0.92 and 0.08, respectively. For the esterase (Es-l) locus, six different phenotypes (AA,BB,CC,AB,BC and AC) were found and presumed to be controlled by three codominant alleles(Es-1^A, Es-1^B and Es-1^C) at a single autosomal locus. The frequencies of B and C alleles were 0.47 and 0.40 and those of these alleles were considerably higher than that of A allele of ES-1. As for the Catalase(Cat) locus, frequency of B allele was showed 0.97, whereas another allele(A) was rarely encounter. In alkalin posphatase(Akp), two phenotype (Akp and akp) were demonstrated and controlled by autosomal dominant(F) and recessive(S) alleles. S allele with frequency of 0.74 showed the higher frequency than that of other allele F in Korean Native Chicken.
In order to find out KNC-specific DNA marker, we performed PCR with 10 oligonucleotide arbitrary primer, which composed of 50-80% G+C content. Of 150 primers(RAPD), we selected 10 primers for polymorphic and reproducible amplification band pattern. Several amplified bands by five primer were specific to KNC. Especially, with primer #327, #399, we was able to identify two bands which appeared only in 6 populations of KNC.
